BET protein inhibitor JQ ⁃1 regulates the differentiation of peripheral blood B cells in systemic lupus erythematosus

Abstract:

Objective To investigate the effect of BET protein inhibitor JQ⁃1 on the differentiation of peripheral blood B cells in systemic lupus erythematosus(SLE). Methods We collected the peripheral blood from 46 SLE patients,from which B cells were isolated,purified and cultured in vitro. Flow cytometry was used to detect the proportion of apoptosis,survival and proliferation of peripheral blood B cells,the proportion of subpopu⁃ lation cells and the proportion of B cells differentiation into plasmablasts in vitro. Results Compared with the con⁃ trol group,the BET protein inhibitor JQ1 had little effect on the early apoptosis,survival and proliferation of the peripheral blood B cells,but it significantly reduced the rates of plasmablasts and plasma cells so as to reduce the proportion of B cells differentiation into plasmablasts in vitro. Conclusion BET protein inhibitor JQ⁃1 can regu⁃ late the differentiation of peripheral plasma blasts into plasma cells in SLE patients,indicating that it may have potential therapeutic effect on SLE.
