Expression of NLRP3 inflammasome and cytokines in patients with syphilis Serofast state

Abstract:

Objective To detect the expression level of the NLRP3 inflammasome and the associated cytokines of syphilis serofast state,and to investigate the effect of NLRP3 inflammasome in the formation of syphilis serofast. Methods In a case⁃control study of 50 syphilis serofast patients and 50 healthy subjects,qRT⁃PCR was used to detect the expression of NLRP3 inflammasome and associated cytokines mRNA in peripheral blood mononu⁃ clear cells(PBMCs)of patients with syphilis serofast state and healthy subjects ,and to detect the levels of cytokines IL⁃1β and IL⁃18 in the serum by ELISA. Results The expressions of NLRP3,ASC,Caspase⁃1,IL⁃1β and IL⁃18 mRNA in PBMCs of patients with syphilis serofast state were significantly lower than those of healthy subjects(all P < 0.05),and the contents of IL⁃1β and IL⁃18 in serum of syphilis serofast state patients were also lower than those of healthy controls(both P < 0.05). Conclusion The expression of NLRP3 inflammasome and downstream cytokines decreases in patients of syphilis serofast ,and suggests that it maybe related to the natu⁃ral immunity abnormality of these patients.
