Role of miR ⁃488⁃3p in invasion and migration of trophoblast cells in preeclampsia placenta

Abstract:

Objective To explore the role of miR⁃488⁃3p in invasion and migration of placental tropho⁃ blast cells in preeclampsia. Methods Expression of miR⁃488⁃3p in placenta was detected by qRT⁃PCR,and its correlation with the clinical characteristics of patients with preeclampsia was analyzed. Human chorionic trophoblast cells were cultured in vitro and transfected with miR⁃488⁃3p mimic and inhibitor;Transwell was applied to observe the effect of miR ⁃488⁃3p on trophoblast cell invasion and migration. Bioinformatics software predicted the down⁃ stream targets of miR⁃488⁃3p and verified its expression in placenta;qRT⁃PCR was then used to detect the effect of miR⁃488⁃3p on MMP2 expression in trophoblast cells. Results As compared with that in normal pregnant women placental expression of miR⁃488⁃3p in patients with preeclampsia was significantly increased,and positively corre⁃ lated with systolic blood pressure,diastolic pressure;while expression of MMP2 was decreased. Overexpression of miR⁃488⁃3p inhibited trophoblast invasion and migration,MMP2 expression decreased;while inhibition of miR⁃488⁃ 3p expression developed a contrary result. Conclusions miR⁃488⁃3p increases in the placenta of patients with pre⁃ eclampsia,which may be involved in the pathogenesis of preeclampsia by silencing MMP2 to inhibit invasion and migration of trophoblast cells.
