1,25⁃二羟维生素D3通过上调Nrf2表达抑制内质 网应激改善小鼠中性粒细胞性哮喘的研究

1,25 ⁃Dihydroxyvitamin D3 ameliorates neutrophilic asthma in mice and inhibits endoplasmic reticulum stress by upregulating Nrf2 expression

摘要:

目的 探讨 1,25⁃二羟维生素 D3[1,25(OH)2D3]对小鼠中性粒细胞性哮喘治疗作用以及对 Nrf2 表达、内质网应激状态的影响。方法 SPF Balb/c 小鼠 24 只,雌性,随机数字表法分为 3 组:正常对 照组(A)、中性粒细胞性哮喘模型组(B)、1,25(OH)2D3治疗组(C),每组各 8 只。用 CFA+OVA(完全弗氏 佐剂+鸡卵清蛋白)诱导的方法制作小鼠中性粒细胞性哮喘模型。C 组在每次 OVA 鼻腔滴入激发前给予 0.1% 1,25(OH)2D3腹腔注射(4 μg/kg)。进行无创气道反应性检测,收集肺组织标本和支气管肺泡灌洗 液(BALF);进行病理 HE 染色、观察肺组织病理炎症变化;计数 BALF 中炎症细胞及分类,检测 BALF 中细 胞因子 IL⁃4、IL⁃17、IL⁃5、CXCL2、CXCL8 水平;Ly⁃6G 免疫组化染色了解肺组织中性粒细胞浸润;Western blot 测定肺组织 CHOP、GRP78 Nrf2 蛋白表达水平;检测肺组织 GSH、MDA 浓度;RT⁃PCR 检测肺组织 NQO1 mRNA的表达水平。结果 1,25(OH)2D3治疗组BALF中各细胞因子分泌明显低于模型组(P < 0.05), BALF 中性粒细胞比例比模型组降低,肺组织病理染色观察气道炎症可见比模型组炎症表现减轻,AHR 低;与模型组比较,治疗组小鼠肺组织 CHOP、GRP78 蛋白表达水平显著降低,Nrf2 蛋白水平升高,NQO1 mRNA 的表达水平升高(P < 0.05)。结论 1,25⁃二羟维生素 D3 可通过增强 Nrf2 蛋白的表达调节氧化应 激,并抑制内质网应激,减少炎症细胞因子的分泌,从而减轻中性粒细胞性哮喘小鼠气道炎症。

Abstract:

Objective To investigate the therapeutic effect of 1,25⁃dihydroxyvitamin D3 on neutrophilic asthma inmice,as well asits influence on Nrf2 expression and endoplasmic reticulum stress state. Methods Twenty⁃ four female Balb/c mice were randomly divided into 3 groups:normal control group(A),neutrophilic asthma model group(B)and 1,25(OH)2D3 treatment group(C),with eight mice in each group.A mouse model of neutrophilic asthma was established by the method of CFA+OVA(complete Freund′s adjuvant,chicken ovalbumin). Group C was given 0.1% intraperitoneal injection of 1,25(OH)2D3(4 μg·kg⁃1)before each challenge by intranasal instillation of OVA.A non⁃invasive airway reactivity test was performed,as well as lung tissue samples and bronchoalveolar lavage fluid(BALF). The inflammatory changes in lung tissue were observed using pathological HE staining. BALF inflammatory cells were counted and classified,and levels of IL⁃4,IL⁃17,IL⁃5,CXCL2 and CXCL8 were mea⁃ sured. Ly ⁃6G immunohistochemistry staining was performed to better understand neutrophil infiltration in lung tissue. Western blot was used to determine the protein expression of CHOP,GRP78,and Nrf2 in lung tissue. GSH,MDA concentration in lung tissue,and NQO1 mRNA levels were all found to be elevated. Results The levels of various cytokines in BALF in the 1,25(OH)2D3 treatment groupwere significantly lower than in the model group (P < 0.05),as was the proportion of neutrophils in BALF. Lung histology revealed that the treatment group had less airway inflammation than the model group.When compared to the model group,the protein levels of CHOP and GRP78 in the treatment group were significantly lower,the protein level of Nrf2 was higher,and the expression level of NQO1 mRNA was higher(P < 0.05). Conclusion 1,25⁃Dihydroxyvitamin D3 reduces airway inflammation in neutrophilic asthmatic mice by increasing the expression of the Nrf2 protein,inhibiting endoplasmic reticulum stress,and inhibiting the secretion of inflammatory cytokines.
