熟地黄多糖对前列腺癌PC⁃3细胞增殖凋亡的作用及对VEGF/Akt信号通路的影响

Role of rehmannia glutinosa polysaccharide in proliferation and apoptosis of prostate cancer PC ⁃ 3 cells and effect of rehmannia polysaccharide on VEGF/Akt signaling pathway

摘要:

目的 基于血管内皮生长因子(VEGF)/蛋白激酶 B(Akt)信号通路研究熟地黄多糖对前列腺癌 PC⁃3 细胞增殖凋亡的作用。方法 取对数生长期人前列腺癌 PC⁃3 细胞分为空白组、低、中、高剂量 组,分别加入 0、20、40、60 μmol/L 熟地黄多糖培养。四甲基偶氮唑蓝(MTT)法检测人前列腺癌 PC⁃3 细胞 增殖抑制率;Transwell 侵袭实验检测 PC⁃3 细胞侵袭能力;流式细胞仪检测 PC⁃3 细胞凋亡率;Western blot 法检测细胞内 VEGF/Akt 信号通路相关蛋白表达。结果 随着熟地黄多糖处理浓度增加,PC⁃3 细胞增殖 抑制率逐渐升高,且呈时间梯度升高(P < 0.05),PC⁃3 细胞的穿膜细胞数逐渐减少,细胞凋亡率逐渐升高, 血管内皮细胞生长因子受体 2(VEGFR2)、磷酸化磷脂酰肌醇 3⁃激酶(p⁃PI3K)、磷酸化 p⁃Akt 蛋白表达逐渐 下调,均有明显浓度依赖性(P < 0.05)。结论 熟地黄多糖对前列腺癌 PC⁃3 细胞具有抑制其增殖与侵袭、 促进其凋亡的作用,可能是通过抑制VEGF/Akt 信号通路实现的。

Abstract:

Objective To explore the role of rehmannia glutinosa polysaccharide in proliferation and apoptosis of prostate cancer PC ⁃ 3 cells by studying vascular endothelial growth factor(VEGF)/protein kinase B (Akt)signal pathway. Methods PC⁃3 cells in logarithmic growth phase were divided into blank group and low⁃ medium⁃ and high⁃dose groups,rehmannia glutinosa polysaccharides of 0,20,40,or 60 μmol/L were added to culture. MTT assay was used to detect the inhibition rate of PC ⁃ 3 cell proliferation. The invasion ability of PC ⁃ 3 cells was detected by Transwell invasion assay. The apoptosis rate of PC ⁃ 3 cells was detected by flow cytometry. The protein expression of VEGF/Akt signaling pathway was detected by Western blot. Results With an increase in rehmannia glutinosa polysaccharide concentration,the inhibition rate of PC⁃3 cell proliferation increased gradually in a time⁃dependent way(P < 0.05);the number of penetrating cells in PC⁃3 cells decreased gradually and the apoptosis rate increased gradually,vascular endothelial growth factor receptor 2(VEGFR2),phosphorylated phosphatidylinositol 3⁃kinase(p ⁃PI3K)and phosphorylated p ⁃Akt protein were gradually down ⁃ regulated in a concentration⁃dependent manner(P < 0.05). Conclusions Rehmannia glutinosa polysaccharide can inhibit prolif⁃ eration and invasion of prostate cancer PC ⁃3 cells and promote PC ⁃3 apoptosis,which may result from inhibiting VEGF/Akt signaling pathway.
