miR⁃218⁃5p靶向VEGFA 凋亡并抑制迁移基因促进宫颈癌HeLa 细胞

miR ⁃ 218 ⁃ 5p targets VEGFA gene to promotes apoptosis and inhibits migration of cervical cancer HeLa cells

摘要:

目的 探究 miR⁃218⁃5p 靶向 VEGFA 基因对宫颈癌 HeLa 细胞凋亡和迁移的影响。方法 养宫颈癌细胞 HeLa,细胞分为 miR⁃NC 组、miR⁃218⁃5p 组、si⁃NC 组、si⁃VEGFA 组、miR⁃NC+pcDNA⁃VEGFA 组、miR⁃218⁃5p+pcDNA⁃VEGFA 组。实时荧光定量 PCR(RT⁃qPCR)检测 miR⁃218⁃5p VEGFA mRNA 表达 水平;流式细胞术检测细胞凋亡;Transwell 检测细胞迁移;蛋白质印迹(Western blot)法检测细胞 B 细胞淋 巴瘤 2(Bcl⁃2)、Bax、基质金属蛋白酶(MMP2)、MMP9 蛋白表达;双荧光素酶报告实验检测 miR⁃218⁃5p VEGFA的靶向关系。结果 与正常宫颈上皮细胞NC104比较,宫颈癌细胞HeLa、CaSki、C33A、SiHa中miR⁃ 218⁃5p表达降低,VEGFA mRNA表达升高(P < 0.05)。与miR⁃NC组比较,细胞凋亡率升高,细胞迁移数降低 P < 0.05)。与si⁃NC组比较,si⁃VEGFA组细胞凋亡率升高,细胞迁移数降低(P < 0.05)。miR⁃218⁃5p靶向调 VEGFA表达,上调VEGFA表达可以逆转miR⁃218⁃5p过表达对宫颈癌细胞凋亡和迁移的影响(P < 0.05)。 结论 miR⁃218⁃5p靶向VEGFA 基因促进宫颈癌HeLa 细胞凋亡并抑制迁移。

Abstract:

Objective To explore the effect of miR⁃218⁃5p targeting VEGFA gene on apoptosis and migra⁃ tion of cervical cancer HeLa cells. Methods In the study,cervical cancer cells HeLa were cultured,and the cells were divided into miR⁃NC group,miR⁃218⁃5p group,si⁃NC group,si⁃VEGFA group,miR⁃NC+pcDNA⁃VEG⁃ FA group,miR⁃218⁃5p+pcDNA⁃ VEGFA group. Real⁃time fluorescent quantitative PCR(RT⁃qPCR)was used to detect the expression levels of miR⁃218⁃5p and VEGFA mRNA;flow cytometry was used to detect cell apoptosis Transwell was used to detect cell migration;Western blot was used to detect cellular B⁃cell lymphoma 2(Bcl⁃2), Bax,matrix metalloproteinase(MMP2),MMP9 protein expression;dual luciferase reporter experiment to detect the targeting relationship between miR ⁃ 218⁃5p and VEGFA. Results Compared with normal cervical epithelial cells NC104,the expression of miR⁃218 in cervical cancer cells HeLa,CaSki,C33A and SiHa were reduced,and the expression of VEGFA mRNA were increased(P < 0.05). Compared with the miR⁃NC group,the apoptosis rate in the miR⁃218⁃5p group was increased,and the number of cell migration was decreased(P < 0.05). Compared with the si⁃NC group,the apoptosis rate in the si⁃VEGFA group was increased,and the number of cell migration were decreased(P < 0.05). miR⁃218⁃5p targeted the regulation of VEGFA expression,and regulation of VEGFA expression could reverse the effects of miR ⁃ 218⁃5p overexpression on apoptosis and migration of cervical cancer cells(P < 0.05). Conclusion miR⁃218⁃5p targeting VEGFA gene promotes cervical cancer HeLa cell apoptosis and inhibits migration.
