蒙古族瘦素受体、脂联素受体基因多态性与 2型糖尿病交互作用研究

摘要:

目的 探究瘦素受体(LEPR)基因 rs3790433 rs12037879、脂联素受体(ADIPOR1)基因 rs10920533 ADIPOR2 基因 rs11061971 位点多态性与环境因素交互作用对蒙古族 2 型糖尿病的影响。 方法 采用病例⁃对照方法,病例组来自呼和浩特市、锡林浩特市和通辽市的 238 例蒙古族 2 型糖尿病患 者,对照组为 238 例同期健康蒙古族人群。多重高温连接酶检测反应检测基因位点多态性,广义多因素 降维法(GMDR)分析基因多态性与环境因素的交互作用。结果 rs3790433 位点 CC+CT 基因型、rs1106197 位点 AA+AT 基因型为蒙古族 2 型糖尿病“高危险”基因型。GMDR 结果显示,一阶交互最优模型为 rs3790433 与超重/肥胖的交互作用模型(P < 0.05)。结论 LEPR、ADIPOR 基因位点多态性与蒙古族 2 糖尿病易感性相关,LEPR 和ADIPOR2基因与环境的交互作用可能增加蒙古族2型糖尿病的发病风险。

Abstract:

Objective To explore the interaction between genetic susceptibility of T2DM and LEPR gene rs3790433 and rs12037879,ADIPOR1 gene rs10920533,ADIPOR2 gene rs11061971 and environmental factors in Mongolian type 2 diabetes population. Methods A case⁃control study was carried out.The case group consisted of 238 Mongolians with type 2 diabetes from Hohhot,Xilinhot,and Tongliao.238 healthy Mongolians made comprised the control group. Multiple high temperature ligase detection reaction(iMLDR)was used to detect gene single nucleotide polymorphism(SNP). Generalized multi ⁃factor dimensionality reduction(GMDR)was used to analyze the interaction between the gene and environmental factors. Results The genotype CC+CT of rs3790433 and the genotype AA+AT of rs1106197 were“high risk”genotypes of T2DM in the Mongolian population(P > 0.05). GMDR results show that the first ⁃ order interaction optimal model rs3790433,overweight/obesity has interaction (P < 0.05). Conclusion The SNPs in the LEPR,ADIPOR1,and ADIPOR2 genes have been linked to T2DM susceptibility in the Mongolian population. The interaction of the LEPR and ADIPOR2 genes with environmental factors may increase the risk of T2DM in Mongolians.
