Association of genetic variants of super⁃fnhancer region with survival of HBV⁃related hepatocellular carci⁃ noma

目的 探讨超级增强子区域遗传变异与乙型肝炎病毒(HBV)相关肝细胞癌(hepatocellular carcinoma,HCC)术后患者总生存期(overall survival,OS)的关系。方法 2007 7 月至 2017 12 月于 广西医科大学附属肿瘤医院行肝癌切除术的 866 HCC 患者为研究对象,采用 Cox 比例风险模型、Kaplan⁃ Meier 法分析超级增强子区域遗传变异与患者 OS 的关系。结果 rs61805159(G > A)和 rs4791170(G > A A 等位基因均可增加 HCC 患者死亡风险(均 P < 0.001),rs546220(C > A)A 等位基因降低 HCC 患者 死亡风险(P < 0.001)。而且,携带多个风险基因型较携带 0 个风险基因型的患者生存率降低,死亡风险 更大,存在剂量⁃反应关系(Ptrend < 0.001)。结论 超级增强子区域单个或多个遗传变异位点影响 HCC OS。

Abstract:

Objective To investigate the relationship between genetic variation of super⁃enhancer region and overall survival(OS)of postoperative patients with hepatitis B virus(HBV)related hepatocellular carcinoma (HCC). Methods A total of 866 patients HCC who underwent hepatectomy in Guangxi Medical University Cancer Hospital from July 2007 to December 2017 were studied. The relationship between genetic variation of super enhancer region and OS were analysed using Kaplan⁃Meier analysis and Cox proportional hazards regression model. Results The rs61805159(G > A)and rs4791170(G > A)A allele were associated with higher risk of death in patients with HCC(P < 0.001),whereas rs546220(C > A)A allele was associated with a lower risk of death(P < 0.001). Moreover,patients with multiple risk genotypes had a lower survival rate and a higher risk of death mortality risk than those with 0 risk genotypes,and there was a dose⁃response relationship(Ptrend < 0.001). Conclusion Single or multiple genetic variation of loci in the super⁃enhancer region affected OS of patients with HCC.
