Expression of Src in tongue squamous cell carcinoma tissue and its effects on proliferation,migration and invasion of the carcinoma

Abstract:

Objective To detect the expression level of Src protein related to Src signal pathway in tongue squamous cell carcinoma(TSCC)and its role in the proliferation,migration and invasion of TSCC tissues. Methods From February 2012 to February 2019,20 cases of tongue squamous cell carcinoma and their corresponding adjacent tissues diagnosed by surgical resection and pathological examinations at our hospital were retrospectively collected. After paraffin sections were made,mmunohistochemistry was used to examine the Src expressions in the carcinoma and its adjacent tissues. The relationship between Src expression and clinicopathological characteristics was analyzed. shRNA was used to down⁃regulate the expression of SRC in the carcinoma cell line SCC127. Western Blot and RT⁃PCR were used to verify its inhibitory efficiency. CCK8 and Transwell invasion and migration experi⁃ ments were done to detect the effects of Src shRNA interference on cell proliferation ,invasion and migration. Results The expression of Src on the oral cancer tissues was significantly higher than on the adjacent tissues(P < 0.01)and it was significantly related to the patient′s tumor differentiation,T stage,clinical stage,and lymph metastasis(P < 0.05),insignificantly to gender and age(P > 0.05). Down⁃regulated expression of Src in SCC127 by shRNA significantly inhibited the proliferation,invasion and migration of SCC127. Conclusion The protein of Src is highly expressed on TSCC tissues,which affects its poor prognosis in patients with tongue cancer. Down⁃regulating Src can reduce the proliferation,invasion and migration of TSCC,suggesting that it may be a potential target for diagnosis and treatment of tongue cancer.
