Effect of NRG ⁃1/TGF ⁃β1 on cytoskeleton of vascular smooth muscle

Abstract:

Objective To investigate the effect of neuregulin 1/transforming growth factor β1(NRG ⁃1/ TGF⁃β1)on the cytoskeleton of vascular smooth muscle. Methods Blood vessel tissues of mice were taken,and the changes of cell contractile function after NRG ⁃ 1 intervention were analyzed by immunofluorescence method and the expression of NRG⁃1/TGF⁃β1 was analyzed by phalloidin and immunofluorescence co⁃localization,and the GST fusion protein was used for sedimentation. Technical detection and analysis of the co⁃location of NRG⁃1⁃ICD and α⁃actin(α⁃SMA)were performed. Results TGF⁃β1 could up⁃regulate the expression of NRG⁃1 in human aortic vascular smooth muscle cells. Immunofluorescence staining showed that the smooth muscle cell marker protein α⁃SMA and NRG⁃1 were co⁃located;RT⁃PCR and Western blot found that NRG⁃1 mRNA expression existed in rat smooth muscle cells,human smooth muscle cells and human endothelial cells. Conclusion TGF⁃β1 can up⁃regu⁃ late the expression of NRG⁃1 at the transcription and translation levels;in cell contraction,TGF⁃β1 can promote the expression of α⁃SMA and NRG⁃1⁃ICD.
