Abstract:
Objective The study investigated the effect and mechanism of salidroside pretreated ADSCs on promoting skin wound healing of diabetic rats. Methods The ADSCs were isolated and treated with low glucose medium,high glucose medium,salidroside combined with high glucose medium respectively. Cell proliferation, migration and expression of cytokeratin 19(CK19),CD31,α⁃smooth muscle actin(α⁃SMA)in three groups were detected. The diabetic skin wounds rat model was prepared and transplanted with ADSCs or salidroside pretreated ADSCs. The wound healing was observed and the expressions of CoL ⁃Ⅰ,COL ⁃Ⅲ,CK ⁃19,CD31 and α⁃SMA were detected. Results The A490,migration numbers and the expression of CK⁃19,CD31,α⁃SMA of ADSCs in the high glucose group were lower than those in control group(P < 0.05);the A490,migration numbers and the expression of CK⁃19,CD31,α⁃SMA of ADSCs in high glucose+salidroside group were obviously higher than those in high glucose group(P < 0.05). The wound healing rate and the expression of CoL⁃Ⅰ,COL⁃Ⅲ,CK⁃19,CD31 and α⁃SMA in wound of the ADSCs group and salidroside+ADSCs group were significantly higher than model group (P < 0.05)and the wound healing rate and the expression of CoL ⁃Ⅰ,COL ⁃Ⅲ,CK ⁃19,CD31 and α⁃ SMA in wound of the salidroside+ADSCs group were notable higher than those in ADSCs group(P < 0.05). Conclusion Salidroside pretreated ADSCs could promote the healing of diabetic skin wounds,and promote the proliferation,
