Therapeutic effect of Hyperoside on mice with collagen⁃induced arthritis

Abstract: Objective To observe the Hyperoside(Hyp)therapeutic effect of collagen ⁃induced arthritis (CIA)in mice. Methods The DBA/1 mice were randomly divided into normal control group(= 10 mice per group)and model group(= 35 mice per group). To induce the CIA model,the model group were given bovine type Ⅱ collagen and complete Freund adjuvant emulsifier on day 0 and day 21,the normal control group was injected with saline. To examine the therapeutic effect of hyperoside,on day 31 arthritic mice were randomly divided into three groups(= 8 mice per group)including the CIA group and hyperoside⁃treated group(25 mg/kg and 50 mg/kg). The measurement of paw thickness and the changes of clinical arthritis index in mice were dynamically observed. On the day 52,the mice were killed,the pathological changes of ankle joint were observed by HE staining. The serum levels of tumor necrosis factor⁃α(TNF⁃α)and interleukin⁃6(IL⁃6)were measured by ELISA. Results CIA model was successfully established,then to measure the paw thickness and clinical arthritis index scores on the day 28. The paw thickness and clinical arthritis index scores were significantly decreased in hypero⁃ side⁃treated group(< 0.01). Cytokines were measured by ELISA,the contents of serum TNF⁃α and IL⁃6 in the hyperoside ⁃treated group were significantly lower than those in the CIA group,with significant statistical signifi⁃ cance. Histopathological observation results showed that compared with the CIA group,the synovial hyperplasia and inflammatory cell infiltration of the ankle joint in the hyperoside ⁃treated group were significantly reduced. Conclusion Hyperoside may have a therapeutic effect on rheumatoid arthritis.
