摘要:

目的 探讨 PYCR1 基因在肝细胞癌(hepatocellular carcinoma,HCC)中的表达及其与临床病 理特征和预后的关系。方法 采用 qRT⁃PCR Western blot 技术对 80 HCC 与癌旁组织的 PYCR1 mRNA 和蛋白表达水平进行检测。采用 Cox 比例风险模型对影响 HCC 患者预后的因素进行单因素、多因素分析。采用 Kaplan⁃Meier Log⁃rank 法进行预后分析。结果 HCC 组中 PYCR1 mRNA 和蛋白相对表达量均 明显高于癌旁对照组(均 P<0.05)。HCC 组织中 PYCR1 表达水平与肿瘤大小、卫星灶、血管侵犯、TNM 期有关(均 P<0.05)。PYCR1 表达水平是影响 HCC 患者 5 年总体生存率和无瘤生存率的独立因素(均 P 0.05)。HCC 组织中 PYCR1 低表达组患者 5 年总体生存率、无瘤生存率均明显高于 PYCR1 高表达组(均 P<0.05)。结论 PYCR1 HCC 组织中高表达,与肿瘤大小、卫星灶、血管侵犯、TNM 分期有关。组织中 PYCR1表达水平与HCC 患者预后关系密切。

Abstract:

Objective To explore the expression of PYCR1 gene in hepatocellular carcinoma(HCC)and its relationship with clinicopathological features and prognosis.Methods Expression levels of PYCR1 mRNA and protein in HCC and adjacent tissues from 80 patients were detected by qRT⁃PCR and Western blot. Cox proportional hazards model was used to analyze the prognostic factors of HCC patients. Kaplan⁃Meier method and the log⁃rank test were used to analyze the prognosis. Results Relative expressions of PYCR1 mRNA and protein in HCC group were higher than that in adjacent tissue group(P < 0.05 for both comparisons). Expression of PYCR1 was correlated with tumor size,satellite lesions,vascular invasion and TNM stage(P < 0.05 for all comparisons). PYCR1 expres⁃ sion level was an independent factor for affecting 5⁃year overall survival rate and disease⁃free survival rate in HCC patients(P < 0.05 for both comparisons). The 5⁃year overall survival rate and disease⁃free survival rate in patients with low expression of PYCR1 in HCC tissues were greater than those in patients with high expression of PYCR1 (P < 0.05 for both comparisons). Conclusions PYCR1 is highly expressed in HCC tissues,which is related with tumor size,satellite lesions,vascular invasion,and TNM stage. Expression of PYCR1 is closely related with the
