蜡样芽孢杆菌调控NF⁃κB信号通路以抑制口腔鳞狀細胞癌细胞增殖、迁移及侵袭

The mechanism of Bacillus cereus inhibiting proliferation,migration and invasion of oral cancer cells through regulating NF⁃κB signaling pathway

摘要:

目的 观察蜡样芽孢杆菌对口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)细胞增殖、迁移及侵袭的影响,并探究其机制。方法 CCK⁃8 法检测 Bacillus 感染后 SCC⁃25 细胞增殖活性,划痕实验及 Transwell 检测蜡样芽孢杆菌感染后 SCC⁃25 细胞迁移及侵袭情况。Western blot 检测NF⁃κB 信号通路表达水平。结果 蜡样芽孢杆菌感染后,SCC⁃25 细胞增殖、迁移及侵袭能力明显低于对照组 P < 0.05),加入 NF⁃κB 抑制剂后,细胞活性增强(P < 0.05)。Western blot 结果显示,蜡样芽孢杆菌与 SCC⁃25 共培养后,pP65、pIκBα IKKγ表达水平增加,NF⁃κB 通路被激活。结论 蜡样芽孢杆菌通过调节 NF⁃κB 通路,抑制OSCC细胞的增殖、迁移及侵袭。

Abstract:

Objective To observe the influences of Bacillus cereus on proliferation and migration of oral squamous cell carcinoma,and investigate the mechanism. Methods CCK8 detected the proliferation of SCC ⁃25 infected by Bacillus. The migration and invasion of SCC25 treated by Bacillus was detected by scratch experiment. The intracellular expression of NF⁃ κB signaling pathway was detected by western blot. Results Compared with control group,the proliferation,migration and invasion of SCC⁃25 were significant declined(P < 0.05). And the activity of SCC⁃25 was increased when treated with NF⁃ κB inhibitor(P < 0.05). The western blot showed that SCC⁃25 infected by Bacillus was contributed to the increasing expression of pP65、pIκBα and IKKγ,and the acti⁃ vation of NF⁃κB pathway. Conclusion Bacillus cereus may inhibit the proliferation,migration and invasion of oral squamous cell carcinoma through NF⁃κB signaling pathway.
