Expression of HO⁃1 mRNA and HO⁃1 in sepsis rats

摘要:

目的 研究脓毒症大鼠肺组织中血红素氧化酶(HO⁃1)和 HO⁃1 mRNA 的表达情况。方法 选取 40 只大鼠,随机分为假手术组 20 只和脓毒症组 20 只;两组分别于术前 20 min 和造模后 2、6 h 12 h 进行断头处死大鼠,并制备 HE 染色光镜病理切片;收集 Western 印迹法 HO1⁃1 蛋白活性水平检测 结果和 Realtime PCR 法对 HO⁃1 mRNA 水平的检测结果;通过大鼠盲肠结扎穿孔(CLP)方法构建脓毒症 模型;免疫组化观察各时间点肺组织形态、HO⁃1 mRNA HO⁃1 蛋白水平的表达情况;观察脓毒症大鼠肺损伤组织形态、HO⁃1 水平变化;分析 HO⁃1 与脓毒症中相应指标及肺功能损伤的关系。结果 假手术组肺组织形态一直处于完整状态,而脓毒症组大鼠肺组织形态在肺组织发生了明显的病理变化;各时 间点脓毒症组大鼠体内的 HO⁃1 印迹颜色均很深,在 12 h 印迹颜色最深,其次为 6、24 h 的颜色最浅; 而假手术组在 3 个时间点的 HO⁃1 印迹颜色均很浅,且与脓毒症组一样在 12 h 印迹颜色最深,其次为 6、24 h 的颜色最浅;假手术组一直保持平稳,未发生明显变化,而脓毒症组 OD 值在 2 h 最低,随后逐渐上升, 经比较 3 个时间点内 OD 差异有统计学意义(P < 0.05);脓毒症组大鼠 OD 值在各时间点中 HO⁃1 mRNA 水平均比假手术组高(P < 0.05)。结论 脓毒症组大鼠肺组织中 HO⁃1 的表达量明显高于假手术组,说明 HO⁃1 对脓毒症患者肺组织的病理形态学变化有明显影响;脓毒症组中肺组织的病理形态学损伤随 着肺组织中 HO⁃1 的表达量的增加而逐渐减轻,说明 HO⁃1 对脓毒症患者的肺组织病理形态有明显的保护作用和改善效果。

Abstract:

Objective To study the expression of heme oxidase(HO⁃1)and HO⁃1 mRNA in the lung tissue of septic rats. Methods Forty rats were selected and randomly divided into a sham operation group of 20 rats and a sepsis group of 20 rats;the two groups were decapitated and sacrificed at 20 min before surgery and 2,6 h and 12 h after modeling. HE stained light microscope pathological sections;collected the detection results of HO1⁃1 protein activity level by Western blotting method and the detection results of HO⁃1 mRNA level by Realtime PCR method;constructed sepsis model by rat cecal ligation and perforation(CLP)method;immune group Observe the lung tissue morphology,HO⁃1mRNA and HO⁃1 protein expression at each time point;observe the lung injury tissue morphology and HO ⁃1 level changes in sepsis rats;analyze the corresponding indexes in HO ⁃1 and sepsis And lung function damage. Results The lung tissue morphology of the sham operation group was always in a com⁃ plete state,while the lung tissue morphology of the sepsis group had obvious pathological changes in the lung tissue;the color of HO⁃1 imprints in the sepsis group was very different at each time point. The color of the blot is darkest at 12 h,followed by the lightest color at 6,24 h;while the color of the HO⁃1 blot at the 3 time points of the sham operation group is very light,and the color of the blot at 12 h is the darkest at 12 h like the sepsis group. Followed by the lightest color at 6 and 24 h;the sham operation group has remained stable without significant changes,while the sepsis group has the lowest OD value at 2 h,and then gradually increases. The difference in OD at the 3 time points is statistically significant significance(P < 0.05);the OD value of rats in the sepsis group was higher than that in the sham operation group at each time point(P < 0.05). Conclusion The expression of HO⁃1 in the lung tissue of the sepsis group was significantly higher than that in the sham operation group,indicat⁃ ing that HO⁃1 has a significant effect on the pathomorphological changes of the lung tissue of patients with sepsis


lung tissue in the sepsis group The pathomorphological damage of the lung tissue gradually reduced with the increase of the expression of HO⁃1 in the lung tissue,indicating that HO⁃1 has a significant protective effect and improvement effect on the pathological morphology of the lung tissue of patients with sepsis.
