Abstract: Objective To study the inhibitional effect of the icariin Ⅱ on epithelial⁃esenchymal transition(EMT)in tongue squamous cell carcinoma(TSCC)and explore its regulation through Notch1/PTEN/FAK signalingpathway. Methods Tca⁃8113 cells in logarithmic phase were randomly divided into blank group and ICSⅡlow,middle,high⁃dose groups. The comparisons were made among the groups in terms of cell proliferation inhibitionrate,cell migration,intervention ability,relative expression of Notch1,PTEN,FAK,p⁃FAK protein of eachgroup. Results The proliferation inhibition rate of ICSⅡ low,medium,high⁃dose groups showed an upward trendat 24,48 and 72 hours(P < 0.05). The mobility and the number of transmembrane cells per field,Notch1 proteinrelative expression and p⁃FAK/FAK of the blank group,ICSⅡ low,medium,high⁃dose groups showed a down⁃ward trend(P < 0.05),of which the PTEN protein relative expression showed an increasing trend(P < 0.05).Conclusion ICSⅡ can inhibit TSCC EMT in a dose⁃ependent manner,which is speculated to be related to thedown⁃regulation of Notch1 expression,up⁃regulation of PTEN expression,and inhibition of FAK phosphorylation.
