Association of characteristics of gut microbiome with gestational diabetes mellitus in pregnant women dur⁃ ing the second trimester of pregnancy

Abstract:

Objective To analyze the characteristics of gut microbiome in pregnant women with gestational diabetes mellitus(GDM)and gestational normal glucose tolerance(GNGT)during the second trimester and its relationship with GDM. Methods A total of 42 pregnant women in the second trimester receiving obstetric exami⁃ nation in our hospital from 2019 to 2020 were selected. Fresh feces were collected on the day of OGTT test. DNA sequencing was performed on the V3⁃V4 variable region of bacterial 16SrRNA using IlluminaHiSeq 2500 platform and bioinformatics analysis was performed. Results At the phylum level,there were differences in intestinal flora composition between the two groups,and the OTUs and diversity of GDM group were higher than that of GNGT group. Bacteroidetes and genera,Firmicutes,Actinomycetes and rumen bacterium genera ⁃UCG014 were correlat⁃ ed with glucose metabolism. The sensitivity and specificity of Bacteroidetes and Bacteria as markers for predicting GDM reached 70% ~ 80%. Conclusions The composition and diversity of intestinal flora in GDM women and GNGT women during the middle trimester of pregnancy were different;the intestinal flora in the middle trimester of pregnancy was correlated with the glucose metabolism index,and Bacteroidetes and Bacteroides could be used as a marker for predicting GDM.
