circTCF25抑制mTOR信号通路对TNF-α诱导血管平滑肌细胞增殖和迁移的影响

Effect of circTCF25 on the proliferation and migration of TNF⁃α induced vascular smooth muscle cells by inhibiting mTOR signaling pathway

摘要:

目的 探讨环状 RNA TCF25(circTCF25)对肿瘤坏死因子α(TNF⁃α)诱导血管平滑肌细胞 A7r5)增殖和迁移的影响和可能机制。方法 TNF⁃α(100 ng/mL)诱导 A7r5 后,RT⁃qPCR 检测 circTCF25 mTOR mRNA p70S6k mRNA 表达,CCK⁃8 法和集落形成实验检测细胞增殖,Transwell 实验检测细胞迁移。将 circTCF25 过表达载体转染 A7r5,检测过表达 circTCF25 TNF⁃α处理的 A7r5 增殖、迁移、mTOR mRNA 和p70S6k mRNA 表达的影响。结果 TNF⁃α诱导后A7r5增殖能力、迁移细胞数显著升高,circTCF25 表达显著降低,mTOR mRNA p70S6k mRNA 表达显著升高(P < 0.05);过表达 circTCF25 后,TNF⁃α诱导的 A7r5增殖能力、迁移细胞数显著降低,mTOR mRNA和p70S6k mRNA表达显著降低(P < 0.05)。抑制mTOR 号通路可减轻过表达 circTCF25 TNF⁃α作用的 A7r5 增殖和迁移的影响(P < 0.05)。结论 circTCF25 抑制TNF⁃α诱导的血管平滑肌细胞增殖和迁移,其机制可能与阻断mTOR 信号通路有关。

Abstract:

Objective To investigate the effect of circular RNA TCF25(circTCF25)on the proliferation and migration of vascular smooth muscle cells(A7r5)induced by tumor necrosis factor α(TNF⁃α)and explore its possible mechanism. Methods After TNF⁃ α(100 ng/mL)induced A7r5,RT⁃qPCR was used to detect the expression of circTCF25,mTOR mRNA and p70S6k mRNA. CCK⁃8 assay and colony formation assay were used to detect cell proliferation,and Transwell assay was used to detect cell migration. A7r5 were transfected with circTCF25 overexpression vector,and the effects of circTCF25 overexpression on the proliferation,migration mTOR mRNA and p70S6k mRNA expression of TNF⁃α⁃treated A7r5 were detected by the above methods. Results After TNF⁃ α induction ,the proliferation ability of A7r5 and the number of migrated cells were significantly increased,the expression of circTCF25 was significantly reduced,and the expression of mTOR mRNA and p70S6k mRNA was significantly increased(P < 0.05). After overexpression of circTCF25,the proliferation ability was significantly impaired and the number of migrating cells was significantly reduced,and the expressions of mTOR mRNA and p70S6k mRNA of A7r5 induced by TNF ⁃ α were significantly reduced(P < 0.05). Inhibition of the mTOR signaling pathway could alleviate the effect of circTCF25 overexpression on the activity and migration of A7r5 induced by TNF⁃α(P < 0.05). Conclusions circTCF25 can inhibit the proliferation and migration of vascu⁃ lar smooth muscle cells induced by TNF⁃α,for it may inhibit mTOR signaling pathway in mechanism.
