circ_0005276靶向miR⁃557调控类风湿关节炎滑膜 成纤维细胞的增殖、迁移和侵袭

circ_0005276 targeted miR⁃557 regulates the proliferation,migration and invasion of rheumatoid arthritis synovial fibroblasts

摘要:

目的 探讨 circ_0005276 靶向 miR⁃557 对类风湿关节炎滑膜成纤维细胞(RA⁃FLS)增殖、 迁移和侵袭的影响。方法 RT⁃qPCR 检测 circ_0005276 miR⁃557 CIA 大鼠滑膜组织中的表达。将 si⁃circ_0005276、si⁃NC、pcDNA、pcDNA⁃circ_0005276、miR⁃NC、miR⁃557模拟物、si⁃circ_0005276+anti⁃miR⁃NC si⁃circ_0005276+anti⁃miR⁃557 分别转染 RA⁃FLS,CCK⁃8、划痕愈合、Transwell 实验用于检测 RA⁃FLS 的活力、 划痕愈合率以及侵袭数。双荧光素酶报告验证 circ_0005276 miR⁃557 的靶向关系。结果 与正常组滑 膜组织比较,CIA 大鼠滑膜组织中 circ_0005276 表达上调(P < 0.05),miR⁃557 表达下调(P < 0.05)。干扰 circ_0005276 RA⁃FLS 的光密度(OD)值、划痕愈合率、侵袭数显著降低(P < 0.05),miR⁃557 表达升高 P < 0.05)。过表达circ_0005276后RA⁃FLS中miR⁃557表达显著降低(P < 0.05)。过表达miR⁃557后RA⁃FLS OD 值、划痕愈合率、侵袭数显著降低(P < 0.05)。下调 miR⁃557 表达显著减弱了干扰 circ_0005276 表达 RA⁃FLS OD 值、划痕愈合率和侵袭数的影响(P < 0.05)。结论 干扰 circ_0005276 可通过靶向促进 miR⁃557表达,从而抑制RA⁃FLS 增殖、迁移和侵袭。

Abstract:

Objective To investigate the effect of circ_0005276 targeted miR⁃557 on the proliferation migration and invasion of rheumatoid arthritis synovial fibroblasts(RA⁃FLS). Methods RT⁃qPCR was used to detect the expression of circ_0005276 and miR⁃557 in synovium of CIA rats. Si⁃circ_0005276,si⁃NC,pcDNA pcDNA⁃circ_0005276,miR⁃NC,miR⁃557 mimic,si⁃circ_0005276+anti⁃miR⁃NC and si⁃circ_0005276+anti⁃miR⁃ 557 were transfected into RA⁃FLS,CCK⁃8,scratch healing and Transwell assay to detect the activity,scratch heal⁃ ing rate and invasion number of RA⁃FLS. Double luciferase report verified the targeting relationship between circ_ 0005276 and miR⁃557. Results Compared with the normal synovial tissue,the expression of circ_0005276 in synovial tissue of CIA rats was up⁃regulated(P < 0.05)and the expression of miR⁃557 was down⁃regulated(P < 0.05). After interfering with circ_0005276,the optical density(OD),scratch healing rate and invasion number of RA⁃ FLS decreased significantly(P < 0.05),while the expression of miR⁃557 increased(P < 0.05). The expression of miR⁃557 in RA⁃FLS was significantly decreased after overexpression of circ_0005276(P < 0.05). The OD value scratch healing rate and invasion number of RA ⁃FLS decreased significantly after overexpression of miR ⁃ 557 (P < 0.05). Down⁃regulation of miR⁃557 expression significantly weakened the effect of interfering circ_0005276 expression on the OD value,scratch healing rate and invasion number of RA⁃FLS(P < 0.05). Conclusion Inter⁃ ference with circ_0005276 could promote the expression of miR⁃557 by targeting,thus inhibit the proliferation,mi⁃ gration,and invasion of RA⁃FLS.
