体外光化学疗法对大鼠肾移植抗排斥反应的研究

摘要:

目的 探讨输注体外光化学疗法处理的单核细胞对肾移植大鼠排斥反应影响。方法 首先 手术建立大鼠肾移植模型,分离受体大鼠单个核细胞,制备 8⁃甲氧基补骨脂素联合紫外线光照的大鼠 单核细胞(ECP),根据静脉输注成分不同,将实验分为 4 组(每组 12 只),组 1 接受 ECP 处理,组 2 接受传 统抗排斥药物,组 3 接受安慰剂处理,组 4 非移植组,接受 ECP 组和安慰剂组受体分别于手术前 7 d、手 术当天和术后 7 d 按组别从尾静脉注入 ECP 细胞(ECP 组)或磷酸盐缓冲液 PBS(安慰剂组);接受免疫抑制大鼠术后第 1 天起,皮下注射环孢素 A 5 mg/kg 连续 10 d。组 4 未接受任何处理。观察受体大鼠的淋 巴细胞数、细胞因子变化以及肾功能、移植大鼠肾病理情况。结果 ECP 组和免疫抑制剂组大鼠 IL⁃2、 INF⁃γ 有不同程度减少、IL⁃10 水平升高,其 CD4+ 、CD8+ 均有不同程度下降,血肌酐较安慰剂组低,排斥反应程度较安慰剂轻,与安慰剂组相比差异有统计学意义。结论 ECP 可调节细胞因子和 T 细胞,减轻肾 移植大鼠的排斥反应。

Abstract:

Objective To explore the effect of monocytes treated by extracorporeal photochemotherapy on rejection in rat kidney transplantation. Methods A model of rat kidney transplantation was surgically established. The mononuclear cells from recipient rats were isolated and treated with 8⁃methoxyporalen plus ultraviolet exposure (ECP). The rats were randomly divided into four groups(12 for each group)according to intravenous infusion of different compositions. Group 1 received ECP,group 2 received traditional anti⁃rejection drugs,group 3 received phoaphate buffer solution(PBS,placebo group),and group 4 received no renal transplantation. Group 2 was injected with cyclosporine A of 5 mg/kg subcutaneously from postoperative day 1 for 10 days. The other three groups were injected with ECP,PBS,or normal saline for 7 presurgical days,on the surgical day,and for 7 postsurgical days through the tail vein,respectively. Changes in lymphocyte count,cytokine,renal function and pathology were observed in transplanted mice. Results Levels of IL⁃2,inf⁃γ,CD4+ ,and CD8+ were decreased,while IL⁃10 level increased in groups 1 and 2. The degree of rejection was slighter in groups 1 and 2 than in group 3. There were significant differences among the different groups. Conclusions ECP can regulate cytokines and T cells and reduce rejection of kidney transplantation in rats.
