大果木姜子精油及浸膏对心肌损伤大鼠的保护作用

Protective effect of essential oil and extract from Cinnamomum migao based on myocardial injury in rats

摘要:

目的 探讨大果木姜子精油、浸膏对心肌损伤大鼠的保护作用。方法 将SD大鼠随机分为正 常对照组(CG)、模型组(MG)、复方丹参组(FFDS)、尼可地尔组(Nic),大果木姜子精油低剂量组(GO⁃1)、 精油高剂量组(GO⁃2),浸膏低剂量组(GE⁃1)、浸膏高剂量组(GE⁃2),每组11只。除CG外,各组大鼠冰水力竭游泳14d,第12天后于颈部皮下多点注射异丙肾上腺素(sisoprotereno,ISO)50 mg/kg,连续 3 d,建立心肌 缺血损伤模型。造模后灌胃相应药物,每天 2 次,连续 7 d。采集大鼠治疗前后Ⅱ导联心电图,观察治疗后 HE、Masson 着染的腹主动脉、心肌组织病理变化,试剂盒检测心肌标志物门冬氨酸氨基转移酶(AST)、乳 酸脱氢酶(LDH)、肌酸激酶同工酶(CK⁃MB)、肌钙蛋白Ⅰ(cTn⁃Ⅰ),炎性因子白细胞介素⁃6(IL⁃6)、肿瘤坏 死因子 α(TNF⁃α),和超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。结果 造模大鼠Ⅱ导联心电图 表现主要为室上性心律失常,心肌组织出现炎性细胞浸润,细胞间质发生胶质沉积,说明大鼠心肌损伤模 型构建成功。与 CG 相比,MG 大鼠心电 ST 段抬高,伴有心律失常,心肌组织有大量细胞坏死和炎性细胞 浸润,可见胶原纤维和结缔组织增生;血清中 LDH、AST 活性及 CK⁃MB、cTn⁃Ⅰ、IL⁃6、TNF⁃α 含量显著增高 P < 0.01),SOD 活性降低(P < 0.05),MDA 表达量显著增高(P < 0.05)。与 MG 比较,FFDS、Nic GO⁃1 GO⁃2,GE⁃1、GE⁃2均可明显改善心肌组织、腹主动脉病理损伤及心电变化。相对于GE⁃1、2,GO⁃1更能抑制 cTn⁃Ⅰ、MDA 的表达,GO⁃2 更能抑制 AST、IL⁃6 的水平;相较于 GO⁃1、2,GE⁃1 更能降低 CK⁃MB、TNF⁃α 的含 量,GE⁃2 更能降低 LDH 活性,提高 SOD 活力。结论 大果木姜子精油、浸膏均能通过减轻氧化应激损伤、 抑制心肌损伤标志物表达及炎症反应来修复损伤心肌组织,浸膏部位在降低 LDH、CK⁃MB、TNF⁃α 表达及 提高SOD 活性方面更具优势。

Abstract:

Objective To investigate the protective effects of essential oil and extract of Cinnamomum migao on myocardial injury in rats. Methods SD rats were randomly divided into normal control group(CG), model group(MG),compound salvia miltiorrhiza group(FFDS),nicorandil group(Nic),low dose essential oil group(GO⁃1),high dose essential oil group(GO⁃2),low dose extract group(GE⁃1)and high dose extract group (GE⁃2),with 11 rats in each group. Rats in each group,except those in CG,swam exhaustively with ice water for 14 days,and ISO(50 mg/kg)was injected subcutaneously at multiple points in the neck on the 12th day for consecutive 3 days to establish a myocardial ischemia injury model. After model establishment,the corresponding drugs were intragastrically administered twice a day for 7 days. Lead Ⅱ ECG was collected from the rats before and after treatment to observe the pathological changes of abdominal aorta and myocardium stained with HE and Masson after treatment. Myocardial markers AST,LDH,CK⁃MB,cTn⁃ I,inflammatory cytokines IL⁃6,TNF⁃ α,SOD activity and MDA content were measured by kits. Results The main ECG findings of lead Ⅱ in the model rats were supraventricular arrhythmia,inflammatory cell infiltration in myocardial tissue and glial deposition in interstitial cells,indicating that the rat myocardial injury model was successfully constructed. Compared with those in CG, rats in MG had ST segment elevation,accompanied by arrhythmia,a large number of cell necrosis and inflammatorycell infiltration in myocardial tissue,and collagen fiber and connective tissue proliferation were observed;LDH and AST activities and CK⁃MB,cTn⁃1,IL⁃6,and TNF⁃α contents in serum were significantly higher(< 0.01); SOD activity was decreased(< 0.05),and MDA expression was significantly higher(< 0.05). Compared with those in MG,pathological damage of myocardial tissue,abdominal aorta and ECG changes were significantly improved in FFDS,Nic,GO⁃1,GO⁃2,GE⁃1 and GE⁃2. Compared with that in GE⁃1 and GE⁃1,the expression of cTn⁃1 and MDA in GO⁃1,and the level of AST and IL⁃6 in GO⁃2 were inhibited;the content of CK⁃MB and TNF⁃α in GE ⁃ 1,and the activity of LDH in GE ⁃ 2 were decreased and the activity of SOD was increased. Conclusion Both essential oil and extract of Cinnamomum migao can repair injured myocardial tissue by alleviating oxidative stress injury,inhibiting the expression of myocardial injury markers and inflammatory response,and the extract site has more advantages in reducing the expression of LDH,CK⁃MB,TNF⁃α and increasing the activity of SOD.
