丙泊酚通过下调Yes相关蛋白的表达抑制卵巢癌SKOV3细胞体外的增殖、迁移及侵袭

摘要: 目的 探讨丙泊酚对卵巢癌SKOV3细胞增殖、迁移及侵袭能力的影响。方法 将SKOV3细胞分为空白对照组、溶剂对照组、25μmol/L丙泊酚组、50μmol/L丙泊酚组、100μmol/L丙泊酚组,细胞功能学实验检测丙泊酚对SKOV3 的增殖、迁移及侵袭能力的影响。免疫荧光实验和Western blot实验检测丙泊酚对SKOV3细胞中Yes相关蛋白(yes associated protein,YAP)表达量的影响。建立 YAP 过表 SKOV3细胞系,观察YAP过表达是否干预丙泊酚对SKOV3细胞增殖、迁移及侵袭能力的抑制作用。结果 丙泊酚抑制卵巢癌SKOV3细胞的增殖、迁移及侵袭能力;丙泊酚下调SKOV3细胞核内YAP蛋白表达;YAP过表达干预了丙泊酚对SKOV3细胞增殖、迁移及侵袭能力的抑制作用。结论 在体外,丙泊酚可能通过下调YAP蛋白的表达,抑制SKOV3细胞的增殖、迁移及侵袭能力。

Abstract:

Objective To investigate the effects of propofol on the proliferation,migration and invasion of ovarian cancer SKOV3 cells. Methods SKOV3 cells were divided into blank control group,solvent control group,25 μmol/L propofol group,50 μmol/L propofol group and 100 μmol/L propofol group. Immunofluorescence assay and Western blot were used to detect the effect of propofol on the expression of yes associated protein(Yap in SKOV3 cells. Whether Yap over⁃expression interfered with the inhibitory effect of propofol on proliferation migration and invasion of SKOV3 cells was observed. Results Propofol inhibited the proliferation,migration and invasion of ovarian cancer SKOV3 cells;Propofol down regulated the expression of Yap protein in the nucleus of SKOV3 cells and Yap overexpression interfered with the inhibitory effect of propofol on the proliferation,migration and invasion of SKOV3 cells. Conclusion propofol may inhibit the proliferation,migration and invasion of SKOV3 cells by down regulating the expression of Yap protein.
