摘要:

目的 探讨氨氯地平对去势大鼠前列腺增生的影响及其可能机制。方法 采用 42 只雄性 Sprague⁃Dawley 大鼠(8 月龄),随机分为 6 组,根据每组 7 只的数量建立各分组模型。处死大鼠后,取前列腺腹侧叶并称重,测量各叶体积,计算前列腺指数(PI)。利用苏木精、伊红染色观察组织病理学变化,采用免疫组织化学(IHC)检测原位血管内皮生长因子(VEGF)表达。采用蛋白印迹法检测 Bcl⁃2、Bax 表达水 平,采用 RT⁃PCR 法检测雄激素受体(AR)表达水平。结果 低剂量氨氯地平除降低 Bcl⁃2 的表达水平和 Bcl⁃2/Bax 比值,还能显著降低前列腺组织中VEGF和AR的表达。结论 氨氯地平通过下调VEGF、AR、Bcl⁃ 2和Bcl⁃2/Bax 比值诱导细胞凋亡来抑制前列腺增生。

Abstract:

Objective To explore the influence and mechanism of amlodipine on prostatic hyperplasia. Methods A total of 42 male mice randomly assigned to 6 groups(n = 7 in each group). The rats were sacrificed and the prostatic ventral lobes were removed and weighed. The volume of the lobes was measured to calculate the PI. Hematoxylin and eosin staining were used to observe the histopathological changes,and in situ VEGF expression was evaluated by IHC. Western blot was used to detect Bcl⁃2 and Bax expression levels,and RT⁃PCR was used to detect the AR expression level. Results Low⁃dose amlodipine significantly decreased the VEGF expression in the prostate tissues,in addition to decreasing the Bcl⁃2 levels and the Bcl⁃2/Bax ratio. Conclusions Amlodipine inhibited prostatic hyperplasia by downregulating VEGF,AR,Bcl⁃2 and the ratio of Bcl⁃2/Bax to induce apoptosis.
