摘要:

目的 探讨神经调节蛋白1/转化生长因子β1(NRG⁃1/TGF⁃β1)对血管平滑肌细胞骨架的作用。方法 取小鼠血管组织,采用免疫荧光法对 NRG⁃1 干预后细胞收缩功能变化进行分析,采用鬼笔环肽、免疫荧光共定位分析 NRG⁃1/TGF⁃β1 表达,采用 GST 融合蛋白沉降技术检测分析 NRG⁃1⁃ICD 与α⁃肌动蛋白 α⁃SMA)的共位性。结果 TGF⁃β1在人主动脉血管平滑肌细胞中可上调NRG⁃1表达,经免疫荧光染色可发现平滑肌细胞标志蛋白α⁃SMA NRG⁃1 存在共位性;经 RT⁃PCR Western blot 检测分析发现,在大鼠平滑肌 细胞、人平滑肌细胞以及人内皮细胞中均存在 NRG⁃1 mRNA 表达。结论 在转录和翻译水平上 TGF⁃β1 可上调NRG⁃1的表达;在细胞收缩中,TGF⁃β1对于α⁃SMA与NRG⁃1⁃ICD之间的表达可以起到促进作用。

Abstract:

Objective To investigate the effect of neuregulin 1/transforming growth factor β1(NRG ⁃1/ TGF⁃β1)on the cytoskeleton of vascular smooth muscle. Methods Blood vessel tissues of mice were taken,and the changes of cell contractile function after NRG ⁃ 1 intervention were analyzed by immunofluorescence method and the expression of NRG⁃1/TGF⁃β1 was analyzed by phalloidin and immunofluorescence co⁃localization,and the GST fusion protein was used for sedimentation. Technical detection and analysis of the co⁃location of NRG⁃1⁃ICD and α⁃actin(α⁃SMA)were performed. Results TGF⁃β1 could up⁃regulate the expression of NRG⁃1 in human aortic vascular smooth muscle cells. Immunofluorescence staining showed that the smooth muscle cell marker protein α⁃SMA and NRG⁃1 were co⁃located;RT⁃PCR and Western blot found that NRG⁃1 mRNA expression existed in rat smooth muscle cells,human smooth muscle cells and human endothelial cells. Conclusion TGF⁃β1 can up⁃regu⁃ late the expression of NRG⁃1 at the transcription and translation levels;in cell contraction,TGF⁃β1 can promote the expression of α⁃SMA and NRG⁃1⁃ICD.
