DKK⁃1基因启动子甲基化与强直性脊柱炎病理性成骨的关系

摘要:

目的 探讨 DKK⁃1 基因启动子甲基化与强直性脊柱炎(ankylosing Spondylitis,AS)的病理性 成骨关系。方法 选择 2017 7 月至 2020 1 月于南方医科大学第三附属医院就诊的强直性脊柱炎患 4 例(AS 组)和外伤致股骨颈骨折患者 4 例(FNF 组)为研究对象,Western Blot 检测 DKK⁃1 表达,MSP 检测 DKK⁃1 甲基化状态,qPCR 检测 DNMTs。结果 AS 组较 FNF 组,DKK⁃1 明显降低(P < 0.05),DKK⁃1 基因启动子甲基化水平明显升高(P < 0.05),而 DNMTs 两组比较差异无统计学意义(P > 0.05)。结论 DKK⁃1 基因可能通过高甲基化抑制 DKK⁃1 表达促进 AS 的病理性成骨,可用于评估 AS 的病理性成骨发 展,为 AS 的治疗提供线索。

Abstract:

Objective To investigate the relationship between DKK⁃1 gene promoter methylation and path⁃ ological osteogenesis in Ankylosing Spondylitis(AS). Methods Western Blot was used to detect the expression of DKK⁃1 protein in hip joint capsule tissue from 4 patients with AS and 4 with femoral neck fracture(FNF)caused by trauma in the Third Affiliated Hospital of Southern Medical University. DKK ⁃ 1 gene promoter methylation was examined by Methylation ⁃ specific PCR(MSP). Meanwhile,DNA methyltransferases(DNMTs)were evaluated by Quantitative Real ⁃time polymerase chain reaction(qPCR). Results The expression of DKK ⁃1 protein in AS hip joint capsule tissue was significantly lower than that in FNF group(P < 0.05),while the methylation status of DKK⁃1 gene promoter was significantly higher(P < 0.05). And there was no significant difference about DNMTs between two groups. Conclusion Lower DKK⁃1 promotes the pathological osteogenesis of AS through DKK⁃1 gene promoter hypermethylation,which can be used to evaluate the development of AS pathological osteogenesis and provide clues for the treatment of AS.
