帕瑞昔布钠对神经病理性疼痛大鼠脊髓胶质细胞活化的影响

Effect of parecoxib on spinal glial activation in the rats with neuropathic pain

摘要: 目的 探讨帕瑞昔布钠对早期治疗神经病理性疼痛大鼠脊髓胶质细胞活化的影响。方法 雄性成年SD大鼠50只,随机分为4组:正常对照组(NC组,n = 5)、假手术对照组(SC组,n = 15)、L5脊神经结扎模型组(SNL组,n = 15)、帕瑞昔布钠治疗组(PAR组,n = 15)。PAR组于术后即刻鞘内注射40 μg/μL的帕瑞昔布钠-生理盐水溶液10 μL,SC组和SNL组以等量生理盐水替代。于术前、术后1、3、7 d测量机械缩足反射阈值,并取术后1、3、7 d大鼠腰膨大进行免疫组织化学染色,观察Iba-1和GFAP蛋白表达水平。结果 与 SNL组相比,鞘内注射帕瑞昔布钠后PWT显著上升(P < 0.05),并在第7 d恢复正常;术后1、3、7 d,PAR组Iba-1和GFAP蛋白表达水平均明显低于SNL组(P < 0.05)。结论 术后即刻鞘内注射帕瑞昔布钠可能通过抑制神经病理性疼痛早期脊髓胶质细胞活化,以缓解神经病理性急性期疼痛。

Abstract: Objective To observe the effect of Parecoxib on spinal glial activation in the rats with neuropathic pain. Methods 50 male rats were randomly divided into Normal control group(Group NC, n = 5), Sham control group(Group SC, n = 15), Spinal nerve ligation group(Group SNL, n = 15), Parecoxib treatment group(Group PAR, n = 15). Group PAR recived 10 μL parecoxib saline solution(40 μg/μL) after surgery by intrathecal infusion. SC group and SNL group recived 10 μL saline replacely. Paw withdrawal threshold was assessed by von frey filaments on one day before operation and day 1, 3, 7 after operation for each group, and the changes of IBA-1 and GFAP expression were evaluated by immunehistochemistry on day 1, 3, 7 for each group. Results Compared with SC group, parecoxib not only significantly alleviated SNL-induced mechanical hypersensitivity (P < 0.05), but also reduced the expression of IBA-1 and GFAP protein. Conclusion These Results suggest that intrathecal infusion of Parecoxib can effectively alleviate the acute pain of neuropathic through inhibiting spinal glial activation in the early time of neuropathic pain.
