环状RNA circAAS1对人绒毛膜滋养细胞侵袭及迁移的影响

摘要

目的 探讨 circAAS1 在子痫前期中的表达及对胎盘滋养细胞侵袭迁移的影响。方法 qRT⁃PCR 检测子痫前期组及对照组各 30 例胎盘组织中 circAAS1 的表达,同时与 30 例子痫前期患者临 床指征做相关性分析。采用 Transwell 和划痕实验检测干扰和过表达 circAAS1 对人绒毛膜滋养层细胞 HTR8/SVneo cells)侵袭和迁移的影响;StarBase 软件预测 circAAS1 下游靶点,并在胎盘组织中检测其表 达;最后,qRT⁃PCR 检测滋养细胞中 circAAS1 miR⁃15a⁃5p 表达的影响。结果 与对照组比较,circAAS1 在子痫前期组胎盘组织中表达上调(P < 0.01),并与临床指征呈正相关(P < 0.01)。circAAS1 上调抑制了 滋养细胞的侵袭和迁移(P < 0.05),miR⁃15a⁃5p 表达下调(P < 0.01);而干扰 circAAS1 表达则呈现相反的作 用(P < 0.05)。结论 circAAS1 在子痫前期胎盘组织中高表达并与收缩压及舒张压相关,circAAS1 通过调 miR⁃15a⁃5p 水平抑制滋养细胞行为,提示 circAAS1 可能在滋养细胞的发生、早期诊断和治疗中发挥重 要作用。

Abstract

Objective To investigate the expression of circAAS1 in preeclampsia and its effect on invasion and migration of placental trophoblast cells. Methods qRT⁃PCR was used to detect the expression of circAAS1 in placental tissues of 30 cases of preeclampsia(preeclampsia group)and 30 cases of controls(control group), followed by analyzing the correlation between circAAS1 expression and the clinical indications of the preeclampsia group. The effects of interference and overexpression of circAAS1 on invasion and migration of human chorionic trophoblast cells(HTR8/SVneo cells)were investigated by Transwell and scratch assay. The software of StarBase was used to predict downstream targets of circAAS1 and analyze its expression in placental tissues. Finally,the effect of circAAS1 on miR⁃15a⁃5p expression in the trophoblast cells was detected by qRT⁃PCR. Results Different from the results of the control group,the circAAS1 expression was up ⁃ regulated in the placental tissues of pre⁃ eclampsia group(P < 0.01)and it was positively correlated with clinical indications(P < 0.01). The up⁃regulated circAAS1 inhibited the invasion and migration of trophoblast cells(P < 0.05)so that miR ⁃15a ⁃5p expression was down ⁃ regulated(P < 0.01). Interference with the circAAS1 expression had the opposite effect(P < 0.05). Conclusion circAAS1 is highly expressed in placental tissues of preeclampsia and it is correlated with systolic and diastolic blood pressure. CircAAS1 inhibits trophoblast behavior by regulating miR ⁃ 15a ⁃5p level,suggesting that circAAS1 may play an important role in trophoblast occurrence,early diagnosis and treatment.
