Effects of lnc⁃MEG3 on the proliferation and invasion of pituitary tumor cells by regulating miR⁃10b⁃5p/ CD82 axis

目的 探究 lnc⁃MEG3 对垂体瘤细胞增殖、侵袭的调控机制。方法 取垂体瘤 HP75 细胞分 为对照组、si⁃NC 组、si⁃MEG3 组、pcDNA3.1⁃NC 组、oe⁃MEG3 组、oe⁃MEG3+mimic⁃NC 组、oe⁃MEG3+miR⁃10b⁃ 5p mimic 组。转染后,检测细胞中 lnc⁃MEG3、miR⁃10b⁃5p 和 CD82 mRNA 的表达水平、细胞增殖、迁移、侵袭 能力和细胞中CD82、PCNA、E⁃cadherin、N⁃cadherin、MMP⁃9蛋白表达。结果 Lnc⁃MEG3过表达可降低miR⁃ 10b⁃5p,上调 CD82 和 E⁃cadherin 水平,降低 PCNA、N⁃cadherin、MMP⁃9 水平,抑制垂体瘤细胞增殖、迁移和侵 袭(P < 0.05);上调 miR⁃10b⁃5p 可抑制 CD82,减弱 lnc⁃MEG3 过表达对垂体瘤细胞增殖、侵袭和迁移的抑制作用。双荧光素酶报告基因检测结果显示,转染 miR⁃10b⁃5p mimic 后,细胞中 MEG3⁃WT 和 CD82⁃WT 的荧 光素酶活性显著降低(P < 0.05)。结论 Lnc⁃MEG3 过表达可能通过靶向下调 miR⁃10b⁃5p,进而上调 CD82 的表达,抑制垂体瘤细胞增殖、迁移和侵袭。

Lnc⁃MEG3调控miR⁃10b⁃5p/CD82轴对垂体瘤细胞增殖、侵袭的影响
Objective To explore the regulation mechanism of lnc⁃MEG3 on the proliferation and invasion of pituitary tumor cells. Methods The purchased pituitary tumor HP75 cells were divided into control group,si⁃NC group,si⁃MEG3 silence group,pcDNA3.1⁃NC group,oe⁃MEG3 group,oe⁃MEG3+mimic⁃NC group and oe⁃MEG3+ miR⁃10b⁃5p mimic group. After transfection,RT⁃qPCR was used to detect the expression levels of lnc⁃MEG3,miR⁃ 10b⁃5p and CD82 mRNA of cells in each group. CCK⁃8 method,scratch test and Transwell test were used to detect cell proliferation,migration and invasion abilities. Western blot was used to detect the protein expression of CD82, PCNA,E⁃cadherin,N⁃cadherin,MMP⁃9 of cells. Dual luciferase reporter gene was used to detect the targeting relationship between miR⁃10b⁃5p and lnc⁃MEG3,CD82. Results Overexpression of lnc⁃MEG3 significantly reduced the expression of miR⁃10b⁃5p,up⁃regulated CD82 and E⁃cadherin levels in cells,decreased PCNA,N⁃cadherin, and MMP⁃9 levels,and inhibited the proliferation and migration of pituitary tumor cells and invasion(P < 0.05). Op⁃regulation of miR⁃10b⁃5p inhibited the expression of CD82,and weakened the inhibitory effect of lnc⁃MEG3 overexpression on the proliferation,invasion and migration of pituitary tumor cells. The results of dual luciferase reporter gene detection showed that after transfection of miR⁃10b⁃5p mimic,the luciferase activities of MEG3⁃WT and CD82⁃WT in the cells were significantly reduced(P < 0.05). Conclusion The overexpression of lnc⁃MEG3 may inhibit the proliferation,migration and invasion of pituitary tumor cells by targeting to down⁃regulate miR⁃10b⁃ 5p and then up⁃regulating the expression of CD82.
