摘要:

目的 探讨脂肪间充质干细胞(adipose derived stem cells,ADSCs)及普兰尼克 F127(Pluronic F⁃127,PF⁃127)水凝胶复合物对新西兰大白兔宫腔粘连(intrauterine adhesions,IUA)子宫内膜的修复机制。方法 将雌性普通级新西兰大白兔随机分为 5 组,按照干预治疗方式的不同将其分为假手术组、 IUA 模型组、PF⁃127 水凝胶组、ADSCs 组、ADSCs⁃PF⁃127 水凝胶复合物组,在干预治疗 14 d 后收集兔双侧子宫组织,组织病理学和免疫组织化学方法分析子宫内膜的厚度、腺体数目、纤维化面积比和转化 生长因子⁃β(transforming growth factor beta,TGF⁃β)的免疫表达量变化,以评估子宫内膜增值和纤维化程度。结果 ADSCs⁃PF⁃127 水凝胶复合物组相比 IUA 模型组,其子宫内膜厚度和腺体数目均明显增 高,子宫内膜纤维化面积比和 TGF⁃β 的表达水平均明显下降,而 PF⁃127 水凝胶组和 ADSCs 组与 IUA 型组比较以上病理特征变化不明显。结论 ADSCs⁃PF⁃127 水凝胶复合物在抗黏附和促进子宫内膜再 生的双重修复作用方面具有显著优势,可以作为一种新的治疗策略为未来的基础临床研究提供初步的依据。

Abstract:

Objective To investigate the repair mechanism of adipose ⁃derived stem cells(ADSCs)and Pluronic F⁃127(PF⁃127)complexes on the endometrium of New Zealand white rabbits with intrauterine adhesion (IUA). Methods Ordinary female New Zealand white rabbits were randomly allocated into,according to different intervention methods,sham operation group,IUA model group,PF ⁃127 hydrogel group,ADSCs group,and ADSCs⁃PF⁃127 group. Uterine tissue was bilaterally harvested from the rabbits 14 days following the intervention in the hydrogel complex group. The endometrial thickness,the number of glands,the ratio of fibrosis area and the transforming growth factor ⁃ beta (TGF ⁃ β were analyzed through histopathological and immunohistochemical methods to assess endometrial proliferation and fibrosis. Results Compared with those in the IUA model group the endometrial thickness and the number of glands were significantly increased in the ADSCs ⁃ PF ⁃ 127 hydrogel complex group;the endometrial fibrosis area ratio and TGF⁃β expression were significantly decreased in the ADSCs⁃ PF⁃127 hydrogel group. The pathological characteristics of the PF⁃127 hydrogel group and the ADSCs group did not experience a significant change when compared with that of the IUA model group. Conclusion The ADSCs ⁃PF ⁃ 127 hydrogel complex has marked advantages in the dual repair effect of anti⁃adhesion and promoting endometrial regeneration. This provides a pathway for devising a novel therapeutic regimen to provide reference for future basic clinical studies.
