Association of expression of Tspan ⁃5 at human maternal ⁃ fetal interface with unexplained pregnancy loss

Objective To detect the expression of tetraspanin 5(Tspan⁃5)at human maternal⁃fetal inter⁃ face,to explore the role of Tspan⁃5 in embryo implantation,and to analyze its association with spontaneous abor⁃ tion. Methods A total of 37 patients with missed abortion who were admitted to Guangdong Second Provincial General Hospital were enrolled as the observation group. Meanwhile,age⁃ and gestational age⁃matched women with normal early pregnancy or non⁃pregnant women of child⁃bearing age were selected into the control group. The expression of Tspan⁃5 in the villi,decidua and endometrium was detected by immunohistochemistry. Results The expression levels of Tspan ⁃ 5 in placental villi of women with spontaneous abortion were significantly lower than those in normal early pregnancy group,while its expression levels in decidual tissues were higher than those in nor⁃ mal early pregnancy group. Tspan⁃5 was not expressed in menstrual endometrium of non⁃pregnant women,but was expressed in endometrial stromal cells and glandular epithelial cells in proliferative and secretory phase. The expres⁃ sion levels of Tspan⁃5 in the decidua of women with normal early pregnancy were slightly higher than those in prolif⁃ erative and secretory phase endometrium,but the difference was not statistically significant(F = 1.26,F = 0.296). Conclusion The decreased expression levels of Tspan⁃5 in placental villi of women with spontaneous abortion and insufficient invasion of trophoblasts may be related to the occurrence of spontaneous abortion. Abnormal expression of Tspan⁃5 at the maternal⁃fetal interface is associated with embryo implantation failure or pregnancy loss.
