摘要:

目的 本研究旨在评估输血用浓缩血小板(platelet concentrate,PC)制备和储存过程中表面凝血主要激活因子——组织因子(tissue factor,TF)的表达。方法 在制备当天和储存至 4 d 的同时,通过流式细胞术分析健康的献血员全血和 PC 来源血小板的 TF,同时分析经典的血小板活化标记物,即纤维蛋白原,P⁃选择素和糖蛋白 GPIIb(CD41)。并根据供血者 ABO 血型分析数据。结果 全血中血小板中检测到 TF,在 O 组献血员中发现最高。与全血相比,制备 PC 时血小板表面 TF 更高,在储存过程中进一步增 加。结论 研究结果表明,PC 中血小板表面 TF 得以维持。有必要进行进一步的研究,以研究 TF 水平与 血小板输注接受者的止血反应之间的可能相关性。

Abstract:

Objective To evaluate the expression of tissue factor(TF)on platelet surface during prepara⁃ tion and storage of platelet concentrates(PC)for transfusional use. Methods Platelet TFs in healthy donor whole blood and PC were measured by flow cytometry on the day of preparation and up to 4 days of storage with classical platelet activation markers,i.e.,fibrinogen,P ⁃selectin,and glycoprotein GPIIb(CD41)analyzed as well. The data were analyzed according to blood ABO group. Results TFs were detected on whole blood platelets,with the highest detection ratein O donors. Compared to the whole blood,the detected TFs were higher on the platelet surface,even escalating with the process of storage. The rise in TF levels were positively correlated with the elevations of the other platelet markers. Conclusion Platelet surface TF is contained in PC. Further studies are needed to investigate a possible correlation between the TF levels and the hemostatic response of platelet transfusion recipient.
